Random Mutagenesis
Random mutagenesis is a powerful tool to determine protein structure-function relationships and to improve or alter their characteristics by modifying proteins. Error-prone PCR is a random mutagenesis technique for generating amino acid substitutions in proteins by introducing mutations into a gene during PCR. Mutations are deliberately introduced by error- prone DNA polymerases. After the mutated PCR products are cloned into an expression vector, the resulting mutant library can be screened for changes in protein activity. Due to the lack of structural information, or when such mutations are difficult to predict from protein structure, the Scientists can use random mutagenesis to identify beneficial mutations. Site-Directed Random Mutagenesis is also called Site-Saturation Mutagenesis or Targeted Random Mutagenesis or Domain Mutagenesis. Since conventional random mutagenesis generates large numbers of non-functional mutants rather than functional genes, these approaches inevitably require extensive screening, a highly cost and long time. By targeting specific residues or protein domain, most of codons remain as wild-type, which can make the mutants maintain natural structure of protein backbone despite of hyper diversity of functional region. Therefore, the mutant library can contain high portion of functional mutants that are most likely to yield positive results.
